Estrogen Related Receptor (ERR) Modulators as Novel Potential Agents for Metabolic Disorders

摘要：
The nuclear estrogen-related receptor a (ERRa) plays a central role in the regulation of expression of the genes involved in mitochondrial biogenesis and oxidative metabolism. Small molecular ERRa agonist is considered as novel therapeutic strategy for metabolic disorders. However, the tiny binding pocket of the ERRa ligand binding domain (LBD) makes it “an intractable molecular target”. Herein, we report the identification of the arguably first class of ERRa agonist. The most promising compound DK3 (CAS3) potently and selectively improved the transcriptional functions of ERRa. It also dose-dependently elevated the mRNA levels and the protein levels of ERRa downstream genes both in vitro and in vivo. More importantly, the compound significantly alleviates glucose intolerance and fatty liver symptoms in HFD and Ob/Ob mouse models. While the in vivo efficacies were totally abolished in ERRa knockout mouse model. The further IND enabling study of this compound is undergoing. Our study, for the first time, suggests that ERRa agonist may serve as new therapeutic agent for metabolic symptoms. The agonists of ERRg was also discovered to induce the “Browning” process of white adipose tissue.

主讲人：暨南大学药学院院长丁克教授

个人简介：丁克教授，1995年本科毕业于中国药科大学化学制药专业；1998年获中国药科大学药物化学硕士学位；2001年获复旦大学博士；2001-2005年美国密西根大学从事博士后研究；2005-2006年任密西根大学医学院Research Investigator；2006-2016年任中科院广州生物医药与健康研究院“百人计划”研究员；2016年任暨南大学药学院院长、教授。研究工作重点围绕肿瘤和代谢性疾病等重大临床需求，设计和合成同时具有生物活性和成药性的 “成（类）药性先导化合物”，为创新药物研究奠定基础。近年来成功设计和合成了：1）世界首个亚型选择性ERRa激动剂作为新型抗代谢性疾病药物候选物； 2）具有治疗肿瘤和肺纤维化良好潜力的世界首个选择性DDR1抑制剂；3）具有治疗心肌肥大潜力的世界首个选择性ZAK抑制剂；4）可克服耐药结核菌的新型抗结核分子等成药性先导化合物100余个。
